      分别以生理盐水、柠檬酸钠-甘氨酸液、海水、KCl-NaCl溶液、Hank’s液、D-Hank’s液为稀释液,不同浓度二甲基亚砜(dimethylsulfoxide,DMSO)及海藻糖为抗冻剂,研究超低温冷冻保存香港牡蛎(Crassostrea hongkongensis)精子方法及不同浓度含氨海水对复苏精子的激活效果,以筛选出最适宜精子冷冻保存的稀释液和抗冻剂及其浓度。结果显示,柠檬酸钠-甘氨酸液为稀释液复苏后精子成活率最高,为(82.90%±0.19%);其次是生理盐水(77.57%±0.49%)、D-Hank’s液(72.22%±0.36%)、Hank’s液(70.00%±0.28%)。氨海水终浓度为0.0313%时,复苏后精子活力最强,柠檬酸钠-甘氨酸溶液为稀释液精子活力为(61.25%±3.06%),生理盐水为稀释液精子活力为(68.70%±0.74%)。以生理盐水为稀释液、DMSO浓度为14%冷冻保存时,复苏精子存活率最高,含海藻糖精子存活率为(86.91%±3.14%),不含海藻糖精子存活率为(81.81%±3.68%),以柠檬酸钠-甘氨酸液为稀释液、DMSO浓度为12%时,复苏后精子存活率最高,含海藻糖的为(79.62%±1.95%),不含海藻糖的为(70.58%±2.41%)。

      Normal saline,sodium citrate-glycine,seawater,KCl-NaCl solution,Hank's solution and D-Hank's solution was respectively taken as extender, while different concentrations of DMSO and mycose as cryoprotectant. In order to select the most appropriate sperm cryopreservation extender, cryoprotectant and its concentration,the paper studied the method of spermatozoa cryopreservation of Crassostrea hongkongensis and the activation effect of different concentration ammonia seawater on post-thaw sperm. The results showed that the highest survival rate of post'thaw sperm was 82.90% ±10. 19% with the extender of sodium citrate-glycine,the next was 77. 57 % ±1 0. 49 % with normal saline,72.22% ±10.36% with D-Hank's solution,and 70. 00% ±10.28% with Hank's solution. When the ammonia seawater concentration was 0. 0313%,the post-thaw sperm had the strongest vitality,and the sperm vitality was 61 .25% ±13.06% with the extender of sodium citrate-glycine while 68.70%±10.74% with normal saline. When normal saline was taken as extender and the concentration of DMSO was 14%,the post-thaw sperm had the highest survival rate,and the sperm survival rate was 86. 91%±13.14% or 81.81%±13.68% when mycose was respectively added or wasn't added. When sodium citrate-glycine was taken as extender and the concentrations of DMSO was 12%,post-thaw sperm had the highest survival rate,and the sperm survival rate was 79. 62%± 1 1.95 %or 70. 58% ±12. 41% when mycose was respectively added or wasn't added.
