从齐口裂腹鱼(Sclizothorax prenanti)肠道中分离出菌株Spc0408,采用常规形态特征的观察、生理生化试验、结合16S rDNA基因序列分析并构建系统发育树。结果表明,该菌为革兰氏染色呈阴性,短杆形,无芽孢;该菌理化特性为具有动力性,H2S、尿素阴性、不发酵木糖、鼠李糖、山梨醇等;β-半乳糖苷酶、葡磷胨水、枸橼酸盐阳性;其16S rDNA长度为1443bp,GenBank数据库登录号为JF929912,与GenBank中维氏气单胞菌(Aeromonas veronii)的相似性高达97%;系统发育树显示,菌株Spc0408和多株维氏气单胞菌聚为一簇。综合分离菌的生化特征及分子鉴定结果,判定该菌株是维氏气单胞菌,从而为气单胞菌的分类鉴定提供参考。 |

The purpose of this study was to make a sequence analysis of 16S rDNA genes of Aeromonas veronii isolated from Schizothorax prenanti. Bacterial strain Spc0408 was isolated by morphological characteristics,physiology and biochemistry experiments,together with 16S rDNA sequence analysis,and its phylogenetic tree was established. The result indicated that strain Spc0408 was Gram-negative,rod-shaped without spores; the strain had physical and chemical properties of motility,negative H2S,urea,D-xylose,L-Rhamnose,D-Sorbitol and so forth,positive Citrate,MR-VP, β-galactosidase,etc. 16S rDNA of strain Spc0408 was 1 443 bp in length,and its accession number in GenBank was JF929912 and 16S rDNA exhibited 97% similarity to A. veronii strains from Gen-Bank. Phylogenetic tree revealed that strain Spc0408 and several A. veronii strains gathered into one cluster with98% similarity. The isolated strain was identified as A. veronii based on its biochemical character and molecular identification,which will benefit the classification and identification of Aeromonas of fish. |
