针对大鲵(Andrias davidianus)性腺发育的不同步现象,为提高受精率,对其精液特性和保存条件进行初步研究。人工辅助采集精液,对精液量、酸碱度、密度、活力等基本特性进行常规测定,分别对保护液、光照、温度等保存条件进行了分组研究试验。结果表明,亲鲵的精液量最多可达12mL/kg,pH值6.7~7.0,呈微酸状态;精子密度215万~1075万/mL,精子活力为0.7~0.9。离体保存条件下,室温暗环境(20lx)较光环境(400lx)可延长存活时间0.5h,暗环境低温(0~4℃)较室温(21~22℃)可延长存活时间3d,低温暗环境下原精液中添加保护液比对照组精子存活时间延长6~12h。 |

Gonadal development of Andrias davidanus not synchronized,semen characteristics and preservation conditions of A. davidanus were studied targeting the increase of fertility rate. Semina were collected by artificial assistance approach,and a regular assay was conducted on the basic features such as semen volume,pH,density,vitality,etc. Preservation conditions,namely,protection medium,illumination and temperature,were also studied. The results showed that semen volume of pro-male A. davidanus was up to 12 mL/kg,pH 6. 7一7. 0 of subacid condition ;and that the semen density was(2. 15一10.75)million/mL, sperm motility 0. 7一0. 9. In vitro conservation conditions,dark environment at ambient illuminance (20 lx).The result also indicated that the survival time of the semina was 0.5 h longer than those conserved in the luminous environment (400 lx);low temperature(0-4℃)could extend the survival time of the semina 3d longer than those at room temperature(21一22 ℃)in dark environment ;in the cold dark environment,semina with protection medium added in could extend the survival time 6一12 h longer than the control group. |
