      利用平衡酚-氯仿法分别提取三角帆蚌(Hyriopsis cumingii)斧足、鳃、闭壳肌、性腺、肝脏、心脏中的基因组DNA,采用紫外分光光度测定法、琼脂糖凝胶电泳法、PCR扩增法分析三角帆蚌不同组织基因组DNA的提取质量。结果表明,三角帆蚌6个组织均能成功地提取基因组DNA,但提取的DNA质量存在差异;斧足中提取的DNA纯度、浓度最高,PCR扩增的条带最亮,因而是三角帆蚌提取DNA的最佳组织;其次是肝脏和闭壳肌,心脏和性腺提取的DNA较差。
英文摘要:
      Freshwater pearl mussel,Hyriopsis cumingii,is the most economically important mussel species in China. Extraction of genomic DNA from preserved specimen is a key step to study phylogeny and molecular genetics. DNA extraction from different tissues of H. cumingii was studied and electrophoresis,OD260/OD280,DNA concentration,PCR amplification were used to check the quantity of DNA. This paper shows that the genomic DNA from different tissues of H. cumingii was extracted by saturated phenol-chloroform method. The results indicated that adductor muscle,foot,gill,gonad,liver and heart could be used to extract DNA easily and successfully. In addition,the value of OD260/OD280 for each DNA samples above mentioned was about 1 .8,and the quantity was 10-20 g per 100 mg. The better tissue of extration DNA was heart. Also,clear and bright products of PCR amplification revealed that the DNA might meet the needs of DNA molecular markers.
