      采用组织块移植培养技术,对来源于中华鳖(Trionyx sinesis)肝脏、脾脏、肾脏、心脏、皮肤及裙边组织细胞进行了原代培养,细胞均可从组织块中迁出并生长成单层细胞。对原代培养的单层细胞用胰酶-EDTA消化后传代培养,初步建立了可连续传代的中华鳖肝脏、脾脏、肾脏、心脏及皮肤组织细胞系;还对细胞的培养条件、冷冻保藏与复苏、染色体数目等进行了研究,初步确定中华鳖细胞培养的条件为32℃,MEME或R1640培养基,血清浓度为20%(V/V)。二甲基亚砜(DMSO)冻存保藏后,细胞复苏生长良好,中华鳖细胞染色体数目为2n=66。
英文摘要:
      By using the tissue explantation technique,primary culture of cells derived from liver, spleen,kidney,heart,skin and shell hem tissues of Chinese soft-shelled turtle,Troinyx sinesis,were conducted in vitro. Cells can migrate out around tissues and then outgrow to monolayer in. Continuous cell lines were established from liver spleen,kidney,heart and shell hem after subculture of primary cells with Trypdin-EDTA digestion method. Primary studies on cell culture conditions,cryopreservation and chromosome analysis,etc,were carried out in this paper. The results showed that the optimal conditions for these cell lines are 32℃,MEME or R1640 midium,20 % Defined Fetal Bovine Serum ( DFBS).Cells were cryopreserved with DMSO in liquid nitrogen and grew well after recovery. The chromosome number of cultured kidney cell is 2n=66.
