以拟态弧菌安徽分离株OmpU基因序列为基础,应用DNAStarProtean软件联合采用多种方法对OmpU的二级结构、柔性、亲水性、表面可能性和抗原指数等方面进行分析,预测OmpU的抗原表位,并以此设计多表位疫苗分子序列。结果表明,拟态弧菌OmpU至少含有9个B细胞表位,其中第24~31、56~66、93~111、123~129、
183~198、253~258和275~287共7个区段的平均抗原指数较高。使用AAY接头以epitope5-epitope7-epitope2-epitope6-epitope3-epitope4-epitope1排列方式串联的多表位序列最接近原蛋白构象,各表位间相对独立,抗原性参数也较好,可作为研制多表位疫苗的候选分子序列。

In order to predict the antigenic epitopes in OmpU and design multi - epitope vaccine molecular, the secondary structure ,flexibility, hydrophilicity, sueface probabity and antigenicity index of OmpU were analysed and by DNAStar Protean software basing on OmpU sequence of V. mimicus isolated from Anhui Province. The results showed that there were at least nine B -cell epitopes in OmpU of V. mimicus, the average antigenicity indexs were higher at 24 -31,56 -66,93 -111 ,123 - 129,183 - 198,253 - 258 and 275 - 287 7 domains. Multi - epitope sequence of epitope5 - epitope7 - epitope2 - epitope6 - epitope3 - epitope4 - epitopel in series connected by AAY was the closest to the original sequence of protein conformation, because of the relatively independent of each epitope and the better antigenicity parameters. So, it could be developed as a multi - epitope vaccine candidate molecular. |
