利用特异性引物MHC IIf和MHC IIr,分别从30尾野生中华鲟(Acipenser sinensis)个体、29尾子中华鲟子一代个体和30尾中华鲟子二代个体的基因组DNA中扩增MHC IIB基因的多肽结合位点(PBR)片段,扩增产物长度为168bp。中华鲟野生群体30个样品95个有效克隆中共检测出30条特异序列(单倍型),中华鲟子一代群体29个样品的96个有效克隆中共检测出38条特异序列(单倍型),中华鲟子二代群体30个样品的95个克隆中检测出36条特异序列(单倍型)。单倍型的分布及群体内遗传多样性参数分析表明,中华鲟子一代群体的MHC IIB基因遗传多样性最高,子二代中华鲟群体的MHC IIB基因遗传多样性最低。中华鲟3个群体的非同义替代(dN)与同义替代(dS)比率为1.39、1.48和1.45,非同义替换率均大于同义替换率分析表明正向选择可能是中华鲟MHC IIB多态性的主要因素。该研究结果提供了中华鲟MHC IIB基因的部分遗传信息和遗传变异规律,为中华鲟全人工种群优化和繁殖管理提供理论依据。 |

The peptide binding region (PBR ) of MHC class IIB gene was amplified from 89 individuals which were sampled from 30 wild population, 29 first filial generation progenies and 30 second filial generation progenies of Chinese sturgeon(Acipenser sinensis),using primer pairs of MHC IIf and MHC IIr. Sequence analysis showed that the amplified fragments were 168bp in length. 30 unique sequences(haplotypes)were detected in 95 valid cloning from 30 wild original individuals, 38 haplotypes were detected in 96 valid cloning from 29 individuals of filial generation progenies, and 36 haplotypes were detected in 95 valid clones from 30 individuals of second filial generation progenies. The analysis of distribution of haplotypes and genetic diversity within population showed that the genetic diversity of MHC IIB genes in first filial generation progenies was the most abundant, while the genetic diversity of MHC IIB genes in second filial generation progenies was the lowest. The rates of non-synonymous substitution (dN) were 1.39, 1.48 and 1.45 times higher than the rate of synonymous substitution (dS) in the MHC IIB on three populations, respectively, showing that positive selection may be a major factor of MHC IIB polymorphism. The present results provide primary genetic variation information of Chinese sturgeon MHC IIB genes and it is useful for the Chinese sturgeon artificial population optimization and reproductive management. |
