原癌基因c-fos和c-jun 属于即刻早期基因,在神经元兴奋传导和中枢神经系统活动过程中起着重要作用。通过RACE技术首次克隆了鲢(Hypophthalmichthysmolitrix)肝脏c-fos和c-jun基因,通过生物信息学方法预测分析了该基因的结构和功能。结果表明,鲢肝脏c-foscDNA 全长为1719bp,开放阅读框1059bp,编码352个氨基酸;c-jun cDNA全长1931bp,其中开放阅读框927bp,编码308个氨基酸;二级结构预测可知,c-fos 包含25.85%的α-螺旋,12.78%的延伸链,1.42%的β 折叠和59.94%的无规则卷曲;c-jun包含32.14%的α-螺旋,5.52%的延伸链,1.62%的β-折叠和60.71%的无规则卷曲。分析其功能结构位点,发现在108~172aa 和227~291aa处有1个典型的亮氨酸拉链结构。通过构建氨基酸系统发育树,鲢与草鱼(Ctenopharyngodonidellus)c-fos同源性高达97%,与草鱼c-jun同源性高达95%,而与哺乳动物同源性较低。 |

The proto-oncogene c-fos and c-jun,which are the members of immediate early gene family,play an important role in neurons excitative conduction and in central nervous system activity. The gene c-fos and c-jun in the liver of Hypophthalmichthys molitrix were cloned for the first time by RACE,while the gene's structure and function were predicatively analyzed by bioinformatics method. The entire length of c-fos cDNA was 1 719 bp,which contained a 1 059 bp open reading frame encoding 352 amino acids. The entire length of c-jun cDNA was 1 931 bp,which contained a 927 bp open reading frame encoding 308 amino acids. By the secondary structure forecast,the c fos had 25.85 % of a-helix,12.78 % of extended strand,1.42% of β-sheet,59. 94% of random coil,and the c-jun had 32. 14% of a-helix,5.52% of extended strand,1.62% ofβ-sheet,60. 71% of random coil. By analyzing the function and structure sites of c-fos and c-jun ,a typical leucine zipper motif was respectively found in 108-172 aa and 227-291 aa. By constructing phylogenetic tree of amino acid,it was found that the homology of c-fos between Hypophthalmichthys molitrix and Ctenopharyngodon idellus was as high as 97%,while the homology of c-jun between them was as high as 95 %.But the homology with mammals were low.
