      本文研究了不同浓度亚硝酸盐氮对团头鲂的急性毒性及高铁血红蛋白的影响。实验结果表明:NO2--N对团头鲂幼鱼(体重20.0±1.0 g)24、48、72、96h 的LC50及安全浓度分别为42.2 mg/L、35.9 mg/L、32.5 mg/L、30.1 mg/L及3.0 mg/L。实验鱼在12.0 mg/L、9.0 mg/L、6.0 mg/L、3.0 mg/L的浓度胁迫下,其血液生化指标均发生明显的变化;实验组12.0 mg/L、9.0 mg/L在胁迫24h后白细胞数量、红细胞数量、血细胞比容与对照组相比均差异性显著(P < 0.05)。在胁迫24h后血红蛋白数量与对照组相比均差异性不显著(P > 0.05),但有上升趋势。随着胁迫浓度升高,时间延长,血红蛋白数量降低,高铁血红蛋白比率升高,血红蛋白大量转化为高铁血红蛋白,并且72h后胁迫浓度组6.0 mg/L、9.0 mg/L、12.0 mg/L都与对照组差异性显著(P < 0.05)。本实验为深入了解NO2--N对团头鲂的致毒机理提供基础数据,为养殖环境控制和疾病防控提供基础科学依据。
英文摘要:
      Nitrite is toxic to aquatic organisms, and its toxic levels are species specific. The high protein inputs characteristic of intensive culture may lead to a high nitrite concentration in pond, especially under high regional water temperature, which poses serious threat to healthy aquaculture. This study examined the acute toxicity of nitrite on Megalobrama amhlycephala and its impact on the serum methemoglobin concentration. The adolescent fish (20.0±1.0g) were acclimatized under de-chlorine tapewater, and temperature was set as 26±0.5°C, pH=7.5, DO≥5mg/L. The nitrite concentration in experiment tanks were adjusted with NaNO2. The results showed that the median lethal concentration (LC50) to M. amhlycephala at 24, 48, 72, 96h were 42.2 mg/L, 35.9 mg/L, 32.5 mg/L, 30.1 mg/L respectively, and the safe concentration was 3.0 mg/L. Fish were challenged with 12.0 mg/L, 9.0 mg/L, 6.0 mg/L and 3.0 mg/L, the biochemical parameters of blood in fish were changed obviously. Under the nitrite at 12.0 mg/L or 9.0 mg/L for 24 h, the number of white blood cell (WBC), red blood cell (RBC), and hematocrit changed significant by being compared to the control (p < 0.05), and hemoglobin (Hb) also showed an increasing trend, but without statistical significant level (P > 0.05). After challenge with 12.0 mg/L of nitrite for 24 h, the methemoglobin (MetHb) / hemoglobin (Hb) increased significantly (p < 0.05), and the values increased significantly in fish challenge with 6.0 mg/L, 9.0 mg/L, 12.0 mg/L at 72 h later. The experiment provided a basic data for deeply understanding the mechanism of NO2--N toxicity, served as the scientific basis for the aquaculture environmental control and disease prevention.
