利用PCR测序技术,对采自南四湖和淮河的12个泥鳅(Misgurnusanguillicaudatus)个体的mtDNA控制区3'端416bp序列进行了测序,共发现86个多态性核苷酸变异位点,多态位点比例为0.206;这些核苷酸的变异位点中,55个为插入缺失,31个为碱基替代,变异位点的分布主要集中在6~36、110~158、366~384核苷酸区段中,其余核苷酸区域相对保守。南阳湖、微山湖与淮河3个泥鳅种群间的遗传距离在0.071~0.080,表现出高遗传变异率。 |

The genetic differences of three Misgurnus auguillicaudatus populations from Shandong Nansi Lake and Huaihe River in China were investigated by PCR coupled with sequencing. Sequence comparing showed there were 86 variation loci in the 3’ end 0f 416 bp D-loop region from 12 samples,among which 55 were from insert and deletion and 31 from base substitutions. The proportion of polymorphic loci was 0. 206. Those mutation bases are distributed within nucleotide loci of 6-36 and 110-158 as well 366-384 intensively. Other bases are relatively conservative. The net distances varied from 0. 071 to 0. 080 between groups,which exhibited high mutation rates in loach samples from the above water areas.
