高产壳聚糖酶菌株的筛选及其产酶条件的优化

      从虾塘底泥中分离到1株高产壳聚糖酶的菌株,将其命名为QY01。研究发现其最适培养基组分为胶体壳聚糖1.0%、葡萄糖0.1%、酵母粉0.3%、K2HPO4•3H2O0.2%、MgSO4•7H2O0.1%、NaCl0.5%、(NH4)2SO40.5%、起始pH6。最适培养条件为6%接种量,30℃、160r/min培养72h,在此条件下发酵液酶活可达10.87U/mL,表明筛选到的菌株在降解壳聚糖方面具有较好的利用价值和开发前景。对QY01进行了16SrDNA形态特征及生理生化特性鉴定分析,经序列比对,该菌株与芽孢杆菌属的同源性高达99%;形态特征与生理生化鉴定结果与16SrDNA鉴定结果相符,初步鉴定为芽孢杆菌属(Bacillussp.)。

      A chitosanase-producing bacterium was isolated from soil at the bottom of shrimp pond and named as QY01.The optimum medium composition for producing chitosanase was shown as follows: colloid chitosan 1.0%,glucose 0.1%,yeast extract 0. 3 %,K2HPO4• 3H2O 0.2%,MgSO4.7H2O 0. 1%,MgSO4•7H2O 0. 1%,NaCI 0.5%,(NH4)2SO4 0. 5%,the initial pH 6. The optimal cultivation conditions for QY01 producing chitosanase were 6% amount of inoculation,30℃incubating temperature and shaking cultivation at 160 r/min for 72 hours. The maximal chitosanase activity could reach 10. 87 U/mL when incubated in flask under the oprimal cultural conditions. The present research showed that QY01 has a better value and prospect of development. The strain QY01 was identified 16S rDNA sequence,morphological,biochemical and physiological identification. The sequence data aligned with 16S rDNA sequences deposited in GenBank indicate that the strain QY01 belongs to the genus of Bacillus and the morphological,biochemical and physioogical characteristics also showed that the strain QY01 belongs to Bacillus sp.
