从广西某鱼场的发病罗非鱼(Tilapianilotica)中分离到1株呈β溶血的革兰氏阴性杆菌,在普通琼脂平板上生长,菌落圆形,边缘整齐,灰白色,湿润菌落,直径为0.5~1.0mm,有动力,接触酶阳性,赖氨酸及鸟氨酸脱梭酶阳性,还原硝酸盐为亚硝酸盐,发酵葡萄糖、蔗糖、麦芽糖,经细菌形态、培养特征、生化特性测定结果均符合迟缓爱德华氏菌的特征。应用PCR技术扩增出576bp目的片段,通过PCR产物核苷酸序列测序及Blast比对结果,进一步确定为迟缓爱德华氏菌。致病性试验结果显示分离菌株具有致病性,对氧氟沙星、氯霉素、氟哌酸、恩诺沙星、先锋霉素等高度敏感。

One pathogenic bacterial strain which was gram-negative bacillus with β-hemolysis, was isolated from the sick Tilapia nilotia of the fish farm in Guangxi. The bacterial strain growed on ordinary agar plate, which form a round colony, regular edge, offwhite, wetness and the diameter was 0.5 - 1.0mm, dynamical, positive for con- tact enzyme test, positive for Lysine off-Jun test and Ornithine de-Jun test, reduced nitrate to nitrite. This bacterium ferment glucose, sucrose, maltose. The isolate was Edwarsiella tarda based on the morphology of bacteria, cultural performances, biochemical characteristics. The 576bp fragment was amplified by PCR, and then sequencing and NCBI blasting, the result further indicated that the isolate was Edwardsiella tarda. The virulence of the isolate was approved by the pathogenesis test. The result of the drug sensitivity showed that the isolate has high sensitive to Ofloxacin, Chloramphenicol, Norfloxacin, Enrofloxacin and Cephalosporin. |
