      以胭脂鱼(Myxocyprinus asiaticus)离体血细胞为实验材料,用不同浓度的氯化镉溶液处理。通过瑞士姬姆萨和AO/EB荧光染色,在光学和荧光显微镜下观察了血细胞的形态变化,并统计了细胞的凋亡率。结果显示,在氯化镉的胁迫作用下,胭脂鱼血细胞呈现细胞变圆、核变圆、核边缘化、形成凋亡小体等细胞凋亡的形态特征。较低Cd^2+浓度(0.01mmol/L)胁迫,1h即可产生明显的细胞凋亡诱导效应,并且细胞凋亡率随着Cd^2+浓度的增加和作用时间的延长而升高。表明Cd^2+能诱导胭脂鱼血细胞发生典型的凋亡,而且细胞凋亡率与Cd^2+的浓度和处理时间成明显的剂量-效应关系。

      Effects of Cadmium (Cd^2+) at different concentrations on apoptosis induction in cultured blood cells of Myxocyprinus asiaticus were studied by AO and EB staining for fluorescence observation and Swiss Giemsa staining for optical microscopy observation, The results of morphological characteristics and apoptosis rate analysis showed that Cd^2+ stressed the M. asiaticus blood cells and caused characteristic features of apoptosis, such as rounding of cells and nuclei, condensation and peripherization of nuclear chromatin, and formation of apoptotic bodies. At the low concentration (0.01 mmol/L) of Cd^2+ , apoptosic features were clearly observed in most of M. asiaticus blood cells after 1 h exposure. Rates of apoptosis were elevated with the increase in Cd^2+ concentration and treatment time. All of the results together indicated that apoptosis of M. asiaticus blood cells induced by Cd^2+ is in a doseand time-dependent manner.
